We have shared some common issues regarding to electrophoresis bands before, and we’d like to share some other abnormal phenomena of polyacrylamide gel electrophoresis in other side. We summary these issues for our customers’ reference to find out the reasons and to obtain accurate results and improve the quality of protein separation.
Gel not polymerized Possible
Reasons are:
(a) Insufficient monomer purity, requiring recrystallization.
(b) Dissolved oxygen in the gel solution inhibits gel polymerization. An effective sparging pump should be used.
(c) Ammonium persulfate is ineffective or insufficient in quantity. Freshly prepare or use another batch of ammonium persulfate with higher concentration.
The gel has already polymerized without adding the water layer.
Reasons are:
(a) Cool the gel solution before adding the catalyst to slow down the polymerization rate.
(b) Reduce the amount of TEMED or ammonium persulfate used.
(c) Speed up the operation.
Gel slides out of the electrophoresis chamber after polymerization
This is particularly prone to occur in low-concentration gels. One solution is to wrap a dialysis membrane around the bottom of the tube or use a porous polypropylene base.
Sample not detected after electrophoresis
Reasons are:
(a) Insufficient sample amount. Increase the sample amount.
(b) Staining solution properties or concentration or staining time are insufficient. Replace the staining solution and increase the concentration and staining time.
(c) The sample floated out during loading. Increase the density of the sample solution and handle with care.
(d) The separation gel concentration is too high, and the sample did not enter the gel. Adjust the gel concentration appropriately.
(e) The separation gel concentration is too low, and the sample has been electrophoresed out of the separation gel. Adjust the gel concentration and the optimal electrophoresis conditions.
(f) If the sample is RNA, it may contain proteins that form a large complex that blocks the gel pores. Thoroughly remove the protein from the RNA sample.
(g) The sample contains enzymes that hydrolyze the sample, and the sample degrades during electrophoresis. Thoroughly purify the sample.
