Answer the questions below to determine the most important factors in selecting your power supply.
1.Will the power supply be used for a single technique or multiple techniques?
Consider not only the primary techniques for which the power supply is being purchased,but other techniques you may use in the future. The power supply chosen for submarine gel electrophoresis of DNA may not provide the voltage or current needed for lEF electrophoresis you plan to run in six months. Likewise, the power supply that provides adequate voltage for your 45-50 cm sequencing gels may be insufficient for the 80-100 cm gels you plan to run in the future.
2. Does the power supply provide the required output?
Consider the maximum voltage, current and power requirements. A 2000 volt,100mA power supply may supply adequate voltage for some types of isoelectric focusing, but would not provide sufficient current for other applications such as SDS-PAGE or electroblotting. Also, consider the increased voltage and/or current requirements for running longer gels or multiple gels.
3. Is a constant power, constant current or constant voltage power supply required?
For optimal results,different techniques require different parameters to be held constant during the running. For example, sequencing and isoelectric focusing are best run at constant power, SDS-PAGE and electroblotting are generally run under conditions of constant current and submarine gel electrophoresis of DNS is run at constant voltage. Refer to protocol and manufacturer’s recommendations for each application.
4. Will the power supply be used to run multiple gels or single gels?
As the number of gels running off a single power supply increases current is increased proportionately. For example, single submarine gel may require 100 volts and 75 mA; two gels would require100 volts and 150mA; four gels would require 100 volts and 300mA.
5. Does the power supply have adequate safety features?
This becomes a critical requirement with high voltage power supplies where potentially lethal voltages exist.The power supplies should provide “Shut down-on-cell disconnect” and ground leakage detection to provide adequate protection to the user.
6.What are your country’s electrical requirements?
