Gel Does Not Set Properly
Issue: The gel has patterns or is uneven, especially in high-concentration gels during colder winter temperatures, where the bottom of the separating gel appears wavy.
Solution: Increase the amount of polymerizing agents (TEMED and ammonium persulfate) to speed up the setting process. Ensure that the glass plates are clean to prevent residues from previous gels.
Gel is Fragile
Issue: Low-concentration gels (used for large molecular weight proteins) break during handling.
Solution: Handle the gel gently throughout the process. In colder conditions, slightly increase the amount of polymerizing agents.
Gel Does Not Set
Solution: In low temperatures, increase the amount of polymerizing agents. Ensure that ammonium persulfate is freshly prepared. If the problem persists, re-prepare the buffer solution.
Many Streaks and Smears on Gel After Electrophoresis
Solution: Ensure the gel solution is pure and thoroughly mixed. Control the reuse of electrophoresis buffer or avoid reuse altogether. If economizing on buffer, use fresh buffer in the inner chamber and reused buffer in the outer chamber.
Incorrect Gel Setting Time
Issue: Homemade gels typically set within 30 minutes to an hour, while commercial kits set within 10-20 minutes. If setting is too slow, the polymerizing agent dosage may be insufficient. If setting is too fast, the dosage may be too high, making the gel hard and prone to cracking, and increasing the risk of overheating during electrophoresis.
Solution: Adjust the amount of polymerizing agents based on the actual conditions. The gel setting time is significantly influenced by temperature.
