What is DNA sequencing?
It is a process to determine the exact sequence or order of bases (A,C, G and T) in a DNA molecule. Why do we need to know the sequencing of DNA on certain genes? Here we know some applications. Recently, the technician in our lab to display how to cast gel by our DYCZ-20F electrophoresis apparatus. The technician Mr.Wu shared the ways to inject the gel into the glass room.
The DYCZ-20F electrophoresis apparatus can cast the gel size of 340x450mm. First, we need to prepare all the work we need to cast gel, like gel solution, glass plate, gel bottle, shark teeth comb, spacer(0.4mm),clips.
Before we cast the gel, we need clean and prepare the glass plate first. Here, the experimenter recommended to use water to clean the surface of the glass plates. Then coat the Bind -silane on the surface of the glass plate to help the gel stick on the glass plate, and coat the Repel-silane on the notched glass plate to peel off the gel easily.
The experimenter demonstrated two ways to inject gel solution:
Slightly tilt the gel glass room, and inject the gel solution evenly back and forth. ( Air bubbles should be avoided producing during this process. )
If the experimenter choose this way, he should practice often to master this way.
For beginners, we recommend this way to cast gel. Clap the three sides by clips , and tilt the gel glass room at the angle of 45 degree, then inject the gel solution at your right side until the gel solution filled in the whole gel room.
