The name “Comet Assay” comes from the characteristic comet-like shape that appears in the results: the nucleus of the cell forms the “head,” while the damaged DNA fragments migrate, creating a “tail” resembling a comet.
Principle
The principle of the Comet Assay is based on the migration of DNA fragments in an electric field. Intact DNA remains within the cell nucleus, while damaged or fragmented DNA migrates toward the anode, forming the comet’s “tail.” The length and intensity of the tail are directly proportional to the extent of DNA damage.
Procedure
- Cell Preparation: Cells to be tested are mixed with low-melting-point agarose and spread on microscope slides to form a uniform layer.
- Cell Lysis: The slides are immersed in a lysis solution to remove the cell membrane and nuclear membrane, exposing the DNA.
- Electrophoresis: The slides are placed in an electrophoresis chamber under alkaline or neutral conditions. Damaged DNA fragments migrate toward the positive electrode under the influence of the electric field.
- Staining: After electrophoresis, the slides are stained with a fluorescent dye (e.g., ethidium bromide) to visualize the DNA.
- Microscopic Analysis: Using a fluorescence microscope or specialized software, the comet shapes are analyzed, and parameters such as tail length and intensity are measured.
